Pain involves different regions of the brain, and the rostral anterior cingulate cortex (rACC) helps to process affective pain. One of the pains experienced most by people after surgery is caused by incisions.
Research published in Frontiers in Pharmacology recently evaluated “the effects of CBD , injected either systemically or locally into the rACC, on mechanical allodynia” (Allodynia can lead to the triggering of a pain response from stimuli which do not normally provoke pain) “in a postoperative pain model and on the negative reinforcement produced by relief of spontaneous incision pain. Additionally, we explored whether CBD underlies the reward of pain relief after systemic or rACC injection.”
In this study, male Wistar rats were submitted to a model of incision pain. All rats experienced mechanical allodynia, but it was shown to be less intense after the administration of CBD. In certain doses, when CBD was injected into the rACC, mechanical allodynia was reduced. However, in smaller doses, mechanical allodynia was not reduced, but even then, peripheral nerve block-induced conditioned place preference (CPP). In higher doses, the CPP was completely inverted.
“Additionally, CBD injected systemically or into the rACC at doses that did not change the incision pain evoked by mechanical stimulation significantly produced CPP by itself. Therefore, a non-rewarding dose of CBD in sham-incised rats becomes rewarding in incised rats, presumably because of pain relief or reduction of pain aversiveness.”
The study found that enough evidence was produced to conclude that CBD influences different dimensions of the response of rats to a surgical incision. Also, the rACC was established as a brain area from which CBD can evoke antinociceptive effects in a similar way to the systemic administration. Sensorial and affective dimensions of pain were also found to be modulated by CBD.
